The blaze broke out shortly after 6:00 in the chicken barn on Perth Road 124, north of Milverton.
At one point, fire crews and equipment from Milverton, Shakespeare, Sebringville and Listowel were on the scene. The barn housed close to 13,000 chickens, and all of them were killed.
The cause of the fire remains under investigation.
People in southern Huron and Lambton counties are being told to limit their time outside.
