The crash happened on Wellington Road 7, north of Drayton, at around 4:45 p.m. on Sunday.
The Wellington County OPP, Guelph-Wellington Paramedic Services and local fire departments attended the scene.
Two people were located in the vehicle.
One was transported to a local hospital with serious injuries, and the second occupant was transported to a trauma centre with life-threatening injuries.
People in southern Huron and Lambton counties are being told to limit their time outside.
