Lambton OPP said officers were called to Confederation Line near South Plympton Road at around 4:30 p.m.
The Wyoming resident was taken to hospital but later died as a result of their injuries.
Police said the identity of the driver will not be released.
The area of Confederation Line was closed to traffic for an investigation but reopened shortly after midnight.
People in southern Huron and Lambton counties are being told to limit their time outside.
